Is it possible to define a variable as real, such that

RE(a+b*i)

would give the answer

a

instead of

RE(a) - IM(b)

?
Um... what exactly are you saying? The prime returns whatever value is stored in a when I perform the calculation RE(a+bi), either is CAS or home mode.
I cannot seem to replicate your result "RE(a)-IM(b)"
Wait, I stand corrected. When using x or y instead of a and b, it does give your result, and this is due to how multiplying complex numbers together works; try plugging some numbers in by hand!

Here is the exact answer to your question, step-by-step:

1. Press & release the blue Shift button on your HP Prime.
2. Press & release the CAS button (which takes you to CAS Settings)
3. Ensure there is no checkmark in "Complex"
4. Press & release the CAS button
5. Type or paste in "RE(a+b*i)
6. Press & release the Enter key
7. Displayed result is "a", just as you wanted
